Thymoquinone Ameliorates Acute Kidney Injury Induced by Renal Ischemia-Reperfusion

چکیده

Renal ischemia-reperfusion injury (IRI)is an unavoidable consequence in renal transplantation and multiple clinical settings. A debate has been raised about the particular role of hypoxia-inducible factor (HF-1α) pathogenesis cortex ultrastructural alterations. Also, we investigated antioxidant/anti-inflammatory effect thymoquinone its modulatory on HIF-1α protection against IRI.Adult male Wister albino rats were assigned into 3 groups (n=16); 1) Sham-operated, 2) IRI model 3) pre-treated with 10 mg.kg-1.day-1 (TQ-IRI) for days at reperfusion onset. Following operation, 8 from each group euthanized after hours remaining 24 hours. was assessed by increased blood urea nitrogen, creatinine level, EGTI histological scoreat both 24h. upregulated (p<0.01) correlated tissue reactive oxygen species (ROS) production total oxidant capacity (TAC) consumption. Elevated inflammatory markers (NFkB, MCP-1 VCAM-1) associated IRI.Thymoquinone treatment inhibited accumulation (p<0.01), reduced oxidation/inflammation process markedly diminished injury.

Dexmedetomidine preconditioning ameliorates kidney ischemia-reperfusion injury

Kidney ischemia-reperfusion (I/R) injury is a common cause of acute kidney injury. We tested whether dexmedetomidine (Dex), an alpha2 adrenoceptor (α2-AR) agonist, protects against kidney I/R injury. Sprague-Dawley rats were divided into four groups: (1) Sham-operated group; (2) I/R group (40 min ischemia followed by 24 h reperfusion); (3) I/R group + Dex (1 μg/kg i.v. 60 min before the surgery...

Dexamethasone ameliorates renal ischemia-reperfusion injury.

In the setting of renal ischemia-reperfusion injury (IRI), the effect and mechanism of action of glucocorticoids are not well understood. In rat renal IRI, a single dose of dexamethasone administered before ischemia, or at the onset of reperfusion, ameliorated biochemical and histologic acute kidney injury after 24 h. Dexamethasone upregulated Bcl-xL, downregulated ischemia-induced Bax, inhibit...

Berberine Ameliorates Renal Functional Disorders and Pulmonary Tissue Injury Following Renal Ischemia/Reperfusion in Rats

Introduction: This study investigated the effect of berberine on renal dysfunction and histological damages of the lung induced by renal ischemia/ reperfusion at an early stage. Methods: There were four experimental groups of adult male rats (n=7). Seven days before induction of ischemia, the Ber+I/R group received oral (by gavage) berberine (15 mg/kg/day) while the I/R group received distil...

Amelioration of rat renal ischemia/reperfusion injury by L-Nil

Introduction: Ischemia/reperfusion (IR) injury involves a complex interrelated sequence of events. High levels of nitric oxide (NO) are generated with inducible form of nitric oxide synthase (iNOS) leading to the renal IR injury and glutathione (GSH) depletion. The present study was designed to investigate the effect of L-Nil (N6- (1-Iminoethyl)-L- lysine.hydrochloride), a selective inhibito...

Renal endothelial dysfunction in acute kidney ischemia reperfusion injury.

Acute kidney injury is associated with alterations in vascular tone that contribute to an overall reduction in GFR. Studies in animal models indicate that ischemia triggers alterations in endothelial function that contribute significantly to the overall degree and severity of a kidney injury. Putative mediators of vasoconstriction that may contribute to the initial loss of renal blood flow and ...
